摘 要:目的:探讨化疗药物顺铂、阿霉素的致肝肾损害机制及加味小柴胡汤的保护作用。方法:将40只大鼠随机分为5组,每组8只:空白组每天灌胃生理盐水;顺铂组生理盐水+顺铂(腹腔注射,下同);阿霉素组生理盐水+阿霉素;实验1组加味小柴胡汤+顺铂;实验2组加味小柴胡汤+阿霉素。给药后观察肝肾组织结构、功能及过氧化反应的变化。结果:给大鼠顺铂或阿霉素,皆可导致肝肾功能与结构损害,SOD及GSH-px活性及GSH含量下降,MDA含量升高;二者呈明显正相关,应用加味小柴胡汤则可明显减轻肝肾功能及组织损害,升高T-AOC、SOD、GSH-PX活性及GSH含量,降低MDA含量。结论:顺铂与阿霉素导致的肝肾功能损害与其引发的过氧化反应有关。加味小柴胡汤的肝肾保护作用与其抗氧化作用有关。Objective: To explore the mechanism of hepatic and renal lesion cause by Cisplatin and Adriamycin, and the protective effect of modified radix bupleuri decoction. Methods Totally 40 healthy rats were randomly divided into 5 groups, each had 8 ones. Rats in control group were lavaged with normal saline everday, the other groups were injected intraperitoneally. Cisplatin: group were given normal saline and Cisplatin, Adriamycin group were given normal salin and Adriamycin, experimental group 1 were given modified radix bupleuri decoction and Cisplatin, experimental group 2 were given modified radix bupleuri decoction and Adriamycin. Then observe the changes that occurred on liver and kidney in the aspects of the organizational structre,function and peroxidation. Results After given Cisplatin or Adriamycin, the rats were all got hepatic and renal lesion in fuction and organizational structure, depressed the activity of SOD and GSH-px, degraded the GSH content and increase the level of MDA. Both were positively associated with each other obviously. Modified radix bupleuri decoction could lessen the injury of hepatic and renal function and histology, enhance the activity of T-AOC, SOD, GSH-px and the GSH content, and decrease the level of MDA. Conclusion Cisplatin and Adriamycin-caused hepatic and renal lesion is associated with the peroxidation caused. The protective effect of modified radix bupleueri decoction on hepatic and renal lesion is concered with its antioxidation.
