摘 要:采用急性毒性实验方法研究了不同浓度的甲基绿溶液对水丝蚓急性毒性(LC50),测定了经甲基绿胁迫后水丝蚓超氧化物歧化酶(SOD)的变化.结果表明:(1)甲基绿对水丝蚓24、48、72 h LC50分别为37.6、31.6、29.3 mg/L.(2)24 h实验,28、32、35 mg/L甲基绿溶液对水丝蚓SOD起诱导作用,与对照组比较,32、35 mg/L SOD活性升高,变化显著(P<0.05);40、45、50 mg/L高浓度试液对水丝蚓SOD起抑制作用,与对照组比较,45、50 mg/L动物SOD活性降低,变化显著(P<0.05);(3)48 h实验,28、32、35 mg/L甲基绿溶液对水丝蚓SOD起诱导作用,与对照组相比较,SOD活性升高,变化显著(P<0.05);40、45、50 mg/L高浓度试液对动物SOD起抑制作用,与对照组比较,SOD活性降低,变化显著(P<0.05);(4)72 h实验,水丝蚓SOD活性总体呈下降趋势.其中28 mg/L药剂对动物SOD活性与24 h相比有所升高.作为核染色剂的甲基绿溶液在一定浓度范围内对水丝蚓造成胁迫,影响动物正常行为及生理现象以至危害生命.We use acute poisoning method to study how the different concentration methyl green affects Limnodilus hoffineisteri( LC50 ), and determine how the superoxide dismutase of L. hoffineisteri changing after stressed by methyi green. ( 1 ) We use methyl green to treat L. hoffineisteri in 24, 48, 72h, the LC50 of L. hoffineisteri are relatively 37.6, 31.6, 29.3 mg/L. (2) The experiments of 24h, the 28, 32, 35 mg/L of methyl green inducing the SOD of L. hoffineisteri, comparing with the control group, when in 32, 35 mg/L, the SOD activity increasing , and they have significance of difference ( P 〈 0.05). When in higher concentrations 40, 45, 50 mg/L, the SOD is resisted, comparing with the control group the SOD declines, and they have significance of difference (P 〈0.05). (3) The experiments of 48h, the 28, 32 , 35 mg/L of methyl green inducing the SOD of L. hoffineisteri, comparing with the control group, the SOD activity increases, and they have significance of difference (P 〈 0.05). When in higher concentrations 40, 45, 50 mg/L, the SOD is resisted, comparing with the control group, the SOD activity declines, and they have significance of difference ( P 〈 0.05 ). (4) The experiments of 72h, the SOD activity all decline. Comparing with 24h, the SOD activity is higher in 28 mg/L. The methyl green which is a kind of coloring reagent to nucleus will threaten L. hoffineisteri in determined concentrations and will affect the normal behaviors and even life of animals.
