泻火补肾汤对移植入脑出血大鼠脑内的神经干细胞存活分化的影响

摘　　要：目的:观察泻火补肾中药复方对移植入脑出血大鼠脑内的神经干细胞(neural stem cells,NSCs)存活分化的影响,并探讨其促进被移植干细胞存活的机制。方法:分离新生SD大鼠海马获得NSCs,筛选群体反应性抗体(panel reactive antibody,PRA)阳性SD大鼠并复制脑出血模型,将5-溴脱氧尿嘧啶核苷(5-bromodeoxyuridine,BrdU)标记的NSCs移植入脑出血大鼠脑内。移植灌胃组用泻火补肾汤煎剂灌胃干预,移植组、脑出血组蒸馏水灌胃。用逆转录聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)法检测大鼠脑内干扰素γ(interferon gamma,IFN-γ)、白细胞介素4(interleukin-4,IL-4)mRNA表达;免疫荧光双标记法检测移植NSCs的大鼠脑内BrdU及NF200双阳性细胞。结果:移植灌胃组大鼠脑内IFN-γmRNA的表达下调,而IL-4mRNA的表达增强(P<0.05),BrdU及NF200双阳性细胞增多。结论:泻火补肾方药促进移植的NSCs存活分化可能与诱导IL-4而抑制IFN-γ的表达有关。Objective： To investigate the effects of Xiehuo Bushen Decoction （XHBSD）. a compound Chinese herbal medicine, on the survival and differentiation of transplanted neural stem cells （NSCs） in brains of rats with intracerebral hemorrhage, and to explore the mechanism of Xiehuo Bushen formula in promoting the survival of transplanted NSCs.in the XHBSD group. Rats in the other two groups were administered distilled water. The expressions of interferon gamma （IFN-γ） and interleukin-4 （IL-4） mRNAs were measured by reverse transcription polymerase chain reaction （RT-PCR）; the numbers of BrdU and 200 kD neurofilament （NF200） positive cells were detected by double-labeling immunofluorescence method. Results： The expression of IFN-γ mRNA was down-regulated significantly in the XHBSD group, but the expression of IL-4 mRNA was up-regulated significantly （P〈0.05）. The numbers of BrdU and NF200 positive cells were also increased remarkably in the XHBSD group. Conclusion： XHBSD can promote the survival and differentiation of transplanted NSCs, which may be related to inducing the expression of IL-4 mRNA and inhibiting the expression of IFN-γ mRNA.
