摘  要:目的:通过观察小鼠骨骼肌损伤后修复过程中机械生长因子(MGF)、M-cad基因表达,探讨MGF在伤后修复过程中所起作用;方法:健康雄性C57BL/6小鼠49只,按体重随机分为7组。其中1组不做任何处理作为对照组(C组),另6组均于同一天损伤,即损伤组(injury,1组)。小鼠腓肠肌钝挫伤后不同时期处死取材,对MGF、M-cad基因表达水平做实时定量RT-PCR分析;结果:骨骼肌损伤后1天MGF表达水平即显著升高(P<0.01),第4、第7天持续升高,第7天达峰值,第11天仍显著高于对照组。非损伤腿中MGF表达无显著变化。M-cad在非损伤腿中的表达与损伤腿相似,均在第7天表达达峰值(P<0.01);结论:MGF可能是激活肌卫星细胞的因子之一,但非惟一因子。MGF也可能与多肽合成有关。The purpose of this study was to analyze the MGF'S function in the recover period after muscle injury by investigating the expression of MGF and M-cad. Mice were killed in different time after gastrocnemius injury and MGF and M-cad gene used for RT-PCR quantitative analysis. The result shows that there is a significant increase in MGF mRNA levels after 1 day (P〈0.01) ,which continued increase at 4 and 7 days following injury. It peaks at 7 days, however, MGF' S expression still significant higher than that of control group at 11 days. Compared with the control group, non-injury muscle's MGF mRNA levels have no significant difference. Similar to the injury muscle, M-cad rnRNA levels in contralateral non-injury muscle seemed to peak at 7 days(P〈0.01). It concludes that MGF may be one but not only one factor which can active the rest muscle satellite cells. It may have some relationships with peptide synthesis.
