摘  要:目的:探讨不同频率机械张应变刺激对骨骼肌卫星细胞形态及细胞增殖指标的影响,为寻找适宜的机械刺激负荷提供实验依据。方法:应用Fx-4000T细胞应变加载系统,时体外培养的大鼠骨骼肌卫星细胞施加15%张应变,频率分别为0 Hz、0.5 Hz、1 Hz和2 Hz,加栽时间为每天6 h,连续4 d;以静置培养的卫星细胞作为对照组。采用流式细胞术检测卫星细胞周期变化,采用RT-ICR技术检测不同频率张应变下膏骼肌卫星细胞MGF mRNA表达水平的变化。结果:张应变刺激后的细胞形态为典型长梭形,且排列呈方向性和规律性。结果:与对照组相比,0.5 Hz、1 Hz和2 Hz组的G_0/G_1期细胞数量显著降低,而0.5 Hz和1Hz组的S+G_2/M期细胞数量显著增加(P<0.05);与0 Hz组相比,0.5 Hz、1 Hz和2 Hz组的G_0/G_1期细胞数量显著降低,而S+G_2/M期细胞数量显著增加(P<0.05)。RT-PCR结果显示,与对照组相比,各实验组的MGF mRNA的表达量均增加。但仅有1 Hz组显著高于对照组(P<0.05)。结论:机械张应变刺激促进骨骼肌卫星细胞的增殖能力和MGF mRNA表达,且与张应变刺激的频率有关,其中1Hz的刺激频率最适宜。The purpose of this paper was to study the effects of mechanical stretch frequency on growth of rat musele satellite cells. Skeletal satellite cells were exposed to 15 % elongation of mechanical strain at 0,0. 5,1 and 2 Hz, respectively for 6 hours per day over a period of 4 days using FlexerCell FT-4000T, while static as control. Cell cycle distribution of satellite cells was detected using flow cytometry. Total RNA was extracted from the cells samples and RT-PCR detection was used to evaluate mRNA of MGF relative expression. The result showed that the morphology of satellite cells Became a typical long spindle following mechanical stimulation. Compared with the control group, the amount of cells in G0/C1 phase at 0. 5 Hz, 1 Hz and 2 Hz group was significantly decreased (P〈0.05), while the amount of cells in S+G2/ M phase at 0.5 Hz and 1 Hz increased (P〈0.05),but no at 2 Hz (P〉0.05), Compared with 0 Hz group,the amount of cells in G0/G1 phase at 0. 5 Hz, 1 Hz and 2 Hz group was significantly decreased (p〈0.05). However,the amount of S+G2/M phase cells was significantly increased (P〈 0. 01). The expression of MGF mRNA in 1 Hz group was significantly increased (P〈0.05) when compared with that of the control group, and in other treatment groups increased too, But no significant difference (P〉0. 05). It concluded that the mechanical stretch stimulated the growth of skeletal muscle satellite cells and the expression of MGF mRNA, which was related to the frequency of mechanical strain. The frequency of 1 Hz is the optimum stimulated load.
