益气活血中药对脑出血大鼠脑组织Tie-2受体蛋白表达的影响

摘　　要：目的测定补阳还五汤及其益气组分、活血组分干预后脑出血大鼠脑损伤区组织Tie-2的表达,初步探讨益气活血中药治疗脑出血的作用机制。方法SD大鼠随机分为正常组、假手术组、模型组、益气活血组、益气组和活血组。用Ⅶ型胶原酶诱导脑出血大鼠模型。各组分别灌服相应益气活血法代表方补阳还五汤全方及其益气组分、活血组分药物,于术后1、4、7、14、21和28d分别检测血肿侧脑组织Tie-2的表达。结果正常组、假手术组各时间点Tie-2的表达未见明显阳性信号;模型组、益气组、活血组及益气活血组脑出血后1d即有Tie-2表达,模型组在21d达到高峰、随后下降,益气组、活血组及益气活血组表达高峰时间提前至14d,益气活血组各个时间点表达均高于同时间点其它组。结论益气活血中药可能通过使脑出血血肿周围脑组织Tie-2受体的表达增强,而促进脑出血损伤区血管新生和成熟的作用,并促进脑出血大鼠神经功能的恢复。Objective:To investigate the mechanisms of method of supplementing Qi and activating blood circulation by observing the effects of Buyanghuanwu Decoction,its supplementing Qi and activating blood airculation components on expressions of Tie-2 in brains of rats with intracerebral hemorrhage (ICH).Methods:One hundred and fifty-five SD rats were randomly divided into six groups,including normal group,sham operation gorup,ICH model group,method of supplementing Qi and activating blood circulation treatment group,supplementing Qi treatment group and activating blood circulation treatment group.ICH model was induced by injecting collagenase typeⅦstereotaxically into right globus pallidus.The last 3 groups were administered orally with the classic method of supplementing Qi and activating blood circulation prescription of Buyanghuanwu Decoction,its supplementing Qi and activating blood circulation components respectively .The expressions of Tie-2 were assayed by Western Blotting on the 1st,4th,7th,14th,21st and 28th day after the onset in 5 rats of each group.Results:The expressions of Tie-2 in the model and three treatment groups were observed on the 1st day,the expressions increased gradually,peaked at 21st day in model group and then decreased.The peak time of supplementing Qi treatment group,activating blood cirulation treatment gorup,supplementing Qi and activating blood circulation treatment group was advanced on the 14th day.Tie-2 expressions of supplementing Qi and activating blood circulation treatment group were significantly higher than other groups at every time points.Conclusion:The intervention of expressions of Tie-2 by method of supplementing Qi and activating blood circulation are up-regulated,which may play an important role in the process of angiogenesis following ICH,promoting and microvascular net work reconstruction and functional recovery of the rats' nerve following cerebral hemorrhagae.
