脑出血大鼠心肌组织TNF-α蛋白表达的变化及扶正护脑胶囊的干预作用

摘  要:目的探讨脑出血大鼠心肌组织TNF-α蛋白表达的变化及扶正护脑胶囊的干预作用。方法动物随机分为假手术组、模型组、扶正护脑组(治疗组)和安宫牛黄组(对照组),分别于术后6、24、72三个时相点取材,采用免疫组织化学法测定心肌组织TNF-α蛋白表达,通过图像分析测定IOD值;并与血清CK-MB、cTnI含量作相关性分析。结果TNF-α阳性颗粒主要在心内膜下的心肌细胞和血管内皮细胞的胞浆中表达。结论炎性因子TNF-α参与了脑出血诱发心肌损伤的机制,扶正护脑胶囊通过抑制TNF-α蛋白表达减轻脑出血后心肌的损伤。Objective: To observe the effect of Fuzhenghunao capsule on TNF - α protein expression of myocardial tissues in rats with intracerebral hemorrhage. Methods: 72 rats were randomly divided into 4 groups: sham group, model group, Fuzhenghunao group and Angongniuhuang group. The material was selected at 6, 24, 72h after operation. The protein expression of TNF -α in rat myocardial tissues was detected by the immunohistochemistry method, and measurd the integrated optical density(IOD) with image analysis. Explored the correction between the IOD of TNF -α and the content of CK - MB and cTnI. Results: The positive paritive particles of TNF -α was in the cytosol of myocardial cell and vascular endotheliocyte near to the endocardium. Conclusion: Inflammatory response attends the myocardial damage. Fuzhenghunao capsule can protect the myocardial tissues by inhibiting the protein expression of TNF -α.
