摘  要:光照和黑暗条件下,植物幼苗的形态建成差异十分明显.以双子叶植物大豆和单子叶植物玉米为材料,利用改进的mRNA差异展示技术分析了黄化苗和绿苗的基因表达的差异黄化苗和绿苗基因表达差异可以归纳为:黄化苗基因特异表达,绿苗基因特异表达,黄化苗基因表达增强,绿苗基因表达增强,黄化苗和绿苗基因表达水平相近五种类型.大豆绿荫中基因特异表达类型比黄化苗要多,玉米中则相反,黄化苗基因特异表达类型多于绿苗.试验中还对差异展示技术参数进行厂分析,并建立了回收、扩增和克隆特异PCR片段的方法.讨论了构建光调控或光仰制特异表达基因探针和光形态建成相关基因分离的可行性.Morphogenesis of light-grown plant seedlings differs from that of dark-grown seedlings. A mollified DD-PCR. was used to detecl alternative expressions of genes between etiolated and green seedlings in maize and soybean. Five kinds of alternative expression of genes were founcl by comparing the gene products of seedlings, including light-induced or suljpressed, dark-induced or suppressed genes. Some genes expressed at both light-and darkgrown conditions. It was also found that there were more genes expressed specifically in soybean green seedlings than that in the et iolated. In contrast, more genes expressed in maize etiolated seedlings than that in green seedlings. Procedure of recovering amplified cDNA from sequencing gel w}ls discussed.
