摘  要:本研究运用多种分子生物学方法对母猪胎盘VEGF mRNA全序列进行克隆和分析,结果发现:母猪胎盘VEGFmRNA全长约3500 bp,其完整的cDNA编码190个氨基酸,通过同源性比较预测了VEGF基因的DNA序列全长约为16kb。VEGF基因编码的氨基酸序列与人、小鼠和恒河猴的同源性分别为78.82%、79.44%和96.34%。经生物信息学分析,预测VEGF蛋白理论分子量为22.33 kD,其等电点为7.89;大约在1-20,40-45,50-60,65-80,100-105位之间的氨基酸存在疏水性区域,用TMHMM2.0分析猪VEGF的跨膜结构发现此蛋白所有氨基酸都位于膜表面,证实了VEGF是一种表面蛋白。用signalP 3.0分析发现在1-27位氨基酸可能为信号肽(P=0.999),且可能在26和27位氨基酸之间发生切割(P=0.956)。对VEGF核苷酸序列和氨基酸序列、蛋白质结构和功能的分析,进一步证实本研究得到了VEGF基因mRNA的全序列,为VEGF基因与母猪繁殖性能的关联性研究提供依据。Many molecule biology methods were used to clon and analysis VEGF mRNA sequence of pig,placental in this study.The result showed that the length of the predicted complete mRNA sequen ce of porcine VEGF was 3500 bp,and it coded 190aa.The identities which compared the amino acid sequence to other animals are found high.Compared with the predicted amino acid sequence of human,mouse and monkey,the identic are 78.82%,79.44% and 96.34%,respective-ly.This study predicted that the molecular weight and isoelectric point of Porcine VEGF are 22.33 kD and 7.89;according to hydrophobicity analysis,the position of 1~20,40~45,50~60,65~80,100~105 were hydrophobicity domain;the position of 1~27 maybe a Signal peptide sequence by the signalP 3.0 analysis.And may be it was segmented at postion of between 26 and 27.It's proved that we had obtained the complete porcine VEGF mRNA sequence though above analyses,and the study also predicted that the VEGF is association with Reproductive traits in porcine.
