焦亚硫酸钠对雄性小鼠肝脏Caspase-3和转化生长因子β_1表达的影响

摘　　要：目的研究焦亚硫酸钠对小鼠肝脏中半胱氨酸天门冬氨酸特异性蛋白酶(Caspase-3)、转化生长因子β1(TGF-β1)表达的影响。方法健康雄性昆明小鼠30只随机分为对照组10只和实验组20只,对照组小鼠给予静置自来水饮用;实验组小鼠给予10g.L-1焦亚硫酸钠溶液饮用。分别于第10天和第30天时处死各组小鼠,快速取出肝脏,Zamboni固定液固定,常规冰冻制片,免疫组织化学染色,光镜观察。结果与对照组比较,实验组小鼠肝脏中Caspase-3阳性表达增强(P<0.01),第10天时实验组变化尤为明显;而实验组小鼠肝脏中TGF-β1阳性表达与对照组比较无显著差异(P>0.05)。结论焦亚硫酸钠喂食后可引起小鼠肝脏中Caspase-3表达增高,TGF-β1无明显改变,提示焦亚硫酸钠促使小鼠肝脏中肝细胞凋亡增加。Objective To study the influence of sodium pyrosulfite on the expression of Caspase-3 and tumor growth factor（TGF） -β1 in male mice Liver. Methods Thirty healthy male mice of Kunming Genus were divided into two groups randomly. Ten mice were fed with tap water in control group,and twenty mice were fed with 10 g·L^-1 sodium pyrosulfite in experimental group. On the 10^th day and on the 30^th day, quickly took out Livers of mice, fixed in the Zamboni fixing solution, conventionally produced frozen section and stain section by immunohistochemical technology. Results Compared with the control group,the expression of Caspase-3 in experimental group was increased, especially on the 10^th day. There was no significant difference in the expression of TGF-β1 between two groups. Conclusion Sodium pyrosulfite can increase the expression of Caspase-3, so it clue to that sodium pyrosulfite can increase cell apoptosis of mice Liver.
