摘 要:目的获取扩增片段长度多态性(AFLP)标记,揭示老年性耳聋的发病机制。方法腹腔注射D-半乳糖致衰老豚鼠模型,另加噪音刺激建立豚鼠听力衰退模型,并列模型组和生理盐水对照组中的听力值极端表型个体进行AFLP标记的筛选,并在空白对照组中验证。结果14对选择性扩增引物仅扩增到1个能将两类极端表型绝然区别开来的多态性条带,其预测表型的准确度在83%以上。结论AFLP筛选技术具有较高的功效,所得的标记较以往线粒体标记的有效性高。Objective To screen amplified fragment length polymorphism (AFLP) markers by whole genome scanning on the hearing-loss model of guinea pig and to reveal the molecular mechanism of presbycusis. Methods A hearing loss model on guinea pig was established by D-galactose injection and subsequent noise stimulation. DNA extracted from the inner ear of each guinea pig was subjected to AFLP marker screening. PCR products were applied on the urea-PAGE gel and the polymorphic bands were visualized by silver-staining. AFLP markers were screened in parallel between extreme individuals from the modeled group and control group ,followed by further verification by individuals from the untouched blank group. Resnits Only 1 of the 14 pairs of primers (QT832) discriminated the two extreme groups. The verification result showed that the accuracy rate of discrimination reached up to 83%. Conclusions AFLP screening method is of high efficacy and the marker QT832 can be dependably applied for investigation of hearing deterioration in guinea pig, superior to the efficacy of other PCR-RFLP markers formerly reported.
