摘 要:目的探讨姜黄提取物治疗血管性痴呆(VD)的作用机制,为临床治疗VD提供理论依据。方法采用脑缺血再灌注(I/R)合并尾部放血降压的方法制备VD小鼠模型,利用跳台实验观察小鼠行为学变化,放射免疫学方法检测小鼠血清及脑组织中降钙素基因相关肽(CGRP)和内皮素(ET)含量的变化。结果姜黄提取物可使VD模型小鼠的反应时间缩短,潜伏期延长,错误次数减少,与模型组比较有显著性差异(P<0.05或P<0.01);同模型组相比,姜黄提取物高、中、低剂量组小鼠血清及脑组织中CGRP水平升高,ET水平下降,以高、中剂量组效果显著(P<0.01或P<0.05)。结论姜黄提取物能改善VD模型小鼠学习记忆能力,其作用机制可能与其改变血清及脑组织的ET、CGRP含量,调节两者平衡有关。Objective To investigate the action role of the extractive from curcumin in the treatment of vascular dementia ( VD), to provide the theory bases on the clinical treatment. Methods The mice model of vascular dementia was induced by the method of cerebral ischemia reperfusion and blood pressure reduction through tail bloodletting. The changes in the behavior of the mice model were observed by platform jumping test, and the changes of calcitionin peptide and endothelin in the serum and in the brain tissue were detected by radioimmunoassay. Results The extractive from curcumin shortened the response period, prolonged the latent period and reduced the error times, and there was significant difference between two groups( P 〈0. 05 or P 〈0. 01 ), the ET levels in the serum and in the brain tissue in the extractive from curcumin group of high, middle and low dosage were lower than those in model group, and CGRP levels were higher than that in model group, but the effects of high and middle group were better than those of the latter ( P 〈 0. 01, P 〈 0. 05 ). Conclusions The extractive from curcumin could improve the learning and memorizing abilities in mice model of VD, which might be related to their changing the levels of ET and CGRP and balance between two levels.
