摘 要:针对龙眼组织富含多糖、单宁、色素及酚类物质的特点,采用改良的SDS法和CTAB法进行龙眼顶芽、叶片和茎皮3种组织总RNA的提取与纯化研究。结果表明。两种方法提取的总RNA均具有28、18、5S3条清晰、完整的条带,且无降解现象;A260/A280均介于1.8-2.0之间。A260/A230均大于2.0。说明改良的SDS法和CTAB法提取的总RNA具有很高的纯度。可以满足后续分子生物学研究的要求。Because longan tissues are rich in polysaccharides ,polyphenol compounds, tannin and some other secondary metabolites, the SDS method and CTAB method were improved to conduct extraction and purification of toatal RNA from tissues of longan tissues. The results indicatod that the RNA isolated by the improved RNA isolation method showed clear bands of 28,18 and 5S rRNA,and the value of OD260 nm/ OD280 nm were between in 1.8 and 2.0. These results demonstrate that the quality and purity of the RNA obtained by the two improved RNA isolation methods can meet the demands of molecular biology experiment.
