摘  要:目的建立高效液相色谱法测定人血中磺胺甲噁唑(SMZ)含量的方法。方法采用Hypersil-ODS2不锈钢柱(4.6mm×200mm,5μm),以磷酸二氢钾(0.067mol.L-1)∶乙腈∶甲醇(75∶15∶10,加2%冰醋酸)为流动相,检测波长为240nm,流速为1.2mL.min-1,柱温35℃。结果反相高效液相色谱法测定人血中磺胺甲噁唑(SMZ)的线性范围为10μg^80μg.mL-1,回归方程为A=-3.221+6.180C,r=0.9995。日内差异、日间差异均小于2.5%,平均回收率为100.77%(n=5)。结论该法简便、快速、灵敏,与甲氧苄氨嘧啶(TMP)分离良好,本法适用于磺胺甲噁唑(SMZ)的血药浓度测定。OBJECTIVE To establish a content determination method of Sulfamethoxazole (SMZ) by HPLC. METHODS The chromatographic conditions were as follow: Hypersil-ODS2 column, potassium dihydrogenphosphate(0. 067mol/L)-acetonitrile-methyl alcohol(75 : 15 : 10), take into 2 % acetic acid as mobile phase; UV detector was set at 240nm, flow rate: 1.2mL· min^- 1, column temperature: 35℃. RESULTS The linear range was 10--80ug · mL^-1, regression equation: A = -3. 221 + 6. 180C, r = 0. 9995. RSD 〈 2.5 %, the average recovery of SMZ was 100.77 % (n = 5).CONCLUSION That method is simple, fast, sensitive, separate with Trimethoprim (TMP)good, this method is applicable to determine the blood drug level of SMZ.
