胶束毛细管电泳Sweeping法检测畜肉中残留氟嗪酸

摘　　要：建立胶束毛细管电泳在线Sweeping富集法检测畜肉中残留的痕量氟嗪酸。讨论了缓冲液pH、SDS浓度、进样时间、运行电压、进样压力对富集效果的影响;考察了不同有机溶剂对蛋白的沉淀效果。结果:选择20mmol/L硼砂+80mmol/L十二烷基硫酸钠(SDS,pH=9.6)为缓冲液,电压18kV,进样时间为200s,进样压力15mbar,三氯乙酸为沉淀剂。氟嗪酸富集倍数达500倍,检测限为0.016mg/L,相对标准偏差RSD(n=3)为2.41%。胶束毛细管电泳在线Sweeping富集法可用于畜肉中残留痕量兽药的检测。A micellar electrokinetic chromatography（MEKC）Sweeping technique was developed for the determination of Ofloxacin in beast meat. The effects of the pH of running buffer, the concentration of SDS, injection time,voltage and injection pressure on concentration were discussed. The effect of the different organic solvent on the protein precipitate were studied. Results： A non-coating fused-silica capillary column （48.5 cm×75 μm,effective length 40 cm）was used, 20 mmol/L Na2B4O＋ 80mmol/LSDS（pH9.6）was used as running buffer, the voltage was 18 kV, the injection time was 200 s,the injection pressure was 15 mbar, trichloraacetic acid was used as precipitant. The concentration factor, the detection limit and RSD was 500,0.016 mg/L,2.41%,respectively.The MEKC method based on on-line Sweeping technique can be used to detect the residuals of trace medicines in beast meat.
