摘  要:目的:探讨清热解毒药配伍桔梗汤对内毒素致急性肺损伤模型大鼠肺核因子Kappa B(NF-κB)p65蛋白表达的影响。方法:雄性SD大鼠按随机数字表法分为生理盐水组、急性肺损伤(ALI)模型组、清热解毒药组、清热解毒药+桔梗汤组。给药6天后股静脉注射内毒素脂多糖(LPS)制备ALI大鼠模型,观察各组大鼠肺病理形态学、髓过氧化物酶(MPO)活性及NF-κB p65蛋白表达的变化。结果:与生理盐水组相比,ALI模型大鼠肺组织中性粒细胞(PMNs)浸润明显、毛细血管充血、水肿,肺组织MPO含量及NF-κB p65胞核蛋白表达明显增强(P<0.01或P<0.05),NF-κB p65胞浆表达减弱(P<0.01)。中药干预后肺组织病变明显减轻,肺MPO水平、NF-κB p65胞核蛋白表达也相应下降(P<0.01或P<0.05),且清热解毒药联合应用桔梗汤疗效优于单纯清热解毒中药。结论:桔梗汤对清热解毒药抑制NF-κB p65从胞浆向胞核转移、发挥解毒抗炎功效具有促进作用。Objective:To explore the effect of QingreJiedu herbs in combination with Jiegeng decoction on expression of NF-κB p65 in ALI induced by LPS.Methods:Male Sprague-Dawley rats were randomly divided into normal group, model control group, QingreJiedu group, and the group of QingreJiedu herbs in combination with Jiegeng decoction. The model of ALI was made by injected with a single dose of LPS through vena femorali, the pathologic morphology, MPO activity and the expression of NF-κB p65 was observed.Result: Compared with normal group,infiltration.of PMNs, capillary congestion and swelling were found in model control group ,and the content of MPO and expression of NF-κB p65 in the nuclear were significantly increased (P〈0.01 or P〈0.05), the expression of NF-κB p65 in the cytolymph was decreased(P〈0.01).Both herb groups can decrease the content of MPO and expression of NF-κB p65 in the nuclear (P〈0.01 or P〈0.05),the group of QingreJiedu herbs in combination with Jiegeng decoction is better than QingreJiedu group.Conclusion: Jiegeng decoction can promote the QingreJiedu herbs to inhibit the transfer of NF-κB p65 from endochylema to nuclear ,improve the effect of detoxicating and anti- inflammation.
