摘 要:用动态膜结晶法对溶菌酶结晶进行研究,考察了各种结晶条件对溶菌酶晶形和膜通量的影响,并对结晶条件进行优化,得到了质量较高的溶菌酶晶体.结果表明,当溶菌酶浓度为20-30mg/mL,结晶剂NaCl浓度在40-90g/L,洗脱剂MgCl2浓度在160~250g/L,pH在4.7~8.2范围内,均能得到高质量的溶菌酶晶体.The continuous osmotic membrane crystallization method was adopted to study on the crystallization of lysozyme. The influences of various crystallization conditions on the shape and quality of lysozyme crystals were investigated and some high quality crystals obtained after the crystallization conditions were optimized. The experiments show that high quality lysozyme crystals can be obtained in the following conditions:the concentration of lysozyme was 20--30 mg/mL, 40--90 g/L NaCl as the crystallizing agent, 160--250 g/L MgCl2 as the stripping solutions and the buffer at pH 4.7--8.2.
