摘 要:检测中药复方H3C-1对肝癌细胞SMMC-7721细胞生长,及肝癌相关基因表达的影响。利用MTT比色法检测复方H3C-1对肝癌细胞SMMC-7721细胞增殖方面的影响;利用以肝癌cDNA抑制性消减文库构建的肝癌相关基因微阵列,检测复方H3C-1作用肝癌细胞SMMC-7721 36 h后基因的表达变化。结果显示中药复方H3C-1作用肝癌细胞SMMC-7721后36h,观察到有31个基因表达下调,1个基因上调。中药复方H3C-1对肝癌肿瘤细胞的作用是通过影响多基因的表达改变实现的;该研究为复方H3C-1的抑癌分子机理提供了重要线索。To detect the effects of Traditional Chinese Medicine Compound H3C-1 (TCMC H3C-1) on the growth and gene expression of hepatocarcinoma cell SMMC-7721, the growth effect of hepatocarcinoma cell SMMC-7721 responding to TCMC H3C-1 was detected by MTT (monotetrazoluim), and the gene expression change of hepatocarcinoma cell SMMC-7721 responding to TCMC H3C-1 for 36 hours was explored by microarray of hepatocarcinoma associated genes, which resulted from suppression subtractive hybridization library. After hepatocarcinoma cell SMMC-7721 was treated by TCMC H3C-1 for 36 hours, the analysis of gene expression profiles indicated that 31 genes expression was down-regulated and 1 genes expression was up-regulated. The present study concludes that hepatocarcinoma cells can be affected by TCMC H3C-1 through changes of multi-genes expression. The research provided important clues to the molecular mechanism how hepatocarcinoma cells response to TCMC H3C-1.
