摘　　要：目的分析以羊水细胞进行胎儿ABO血型基因分型及其可行性分析。方法妊娠16周孕妇抽取羊水,从羊水细胞中提取DNA,采用PCR-SSCP技术,通过PCR反应,特异性扩增ABO基因片段,用凝胶电泳检测PCR产物,SSCP硝酸银染色,PCR产物直接测序等分子生物学方法,直接鉴定ABO血型基因型。结果15例高抗体效价的O型孕妇中丈夫A型的9例,B型5例,AB型1例,胎儿羊水细胞基因定型血型与末梢血血型吻合。结论通过羊水细胞,采用PCR-SSCP技术,进行胎儿ABO血型基因定型的方法是可行的。Objective To invbestigate the feasibility of genotyping of ABO blood group in amniocyte.Methods After distilled DNA from women with a gestational period of sixteen weeks and with blood type O.The ABO blood group were identified directly by PCR-SSCP.Results Among fifteen husbands of those pregnant women,9 were with blood type A,five with blood type B,one with blood type AB.The genotyping of ABO blood type detected by PCR-SSCP were coincidence with that detected by using capillary blood.Conclusion It is feasible to detect genotyping of fetal ABO blood group in amniocyte by PCR-SSCP.
