摘  要:小鼠骨肉瘤基因B(Finkef—Biskis—Jinkins murine osteosarcoma B,fosb)是具亮氨酸拉链结构的转录因子激活剂蛋白-1(activator protein-1,AP-1)家族的成员之一。fosb基因在转录拼接过程中会自发地发生剪切截去C端的101个富含脯氨酸的氨基酸区域而形成△fosb,被截断的区域包括转录激活结构域,The recombinant adenoviruses which can overexpress fosb and △2△fosb proteins were produced. Primary calvarial osteoblasts from new born CD1 mice were infected with those adenoviruses by different proportion of Ad-△2△fosb and induced by 5 mmol/L β-glycerophosphate, 50 μg,/ml ascorbic acid and 10 nmol/L dexamethasone for 23 days and 29 days, respectively. The re- sults by Alizarin Red Staining showed that the primary osteoblasts overexpressing △2△fosb protein formed more nodus than the control cells and the nodus in overexpressing △2△fosb protein osteoblasts were smaller than those in control cells on day 23. Differentiation of the primary osteoblasts overexpressing △2△fosb protein increased more than that of the control cells on day 29. It is an evidence for the reason that bone density of △2△fosb transgenic mice was markedly increased throughout the skeleton in new born and mature mice.
