摘 要:目的观察大鼠视网膜缺血-再灌注损伤(retinal ischemial reperfusion injury,RI-RI)后神经调节蛋白-1β(neuregulin-1β,NRG-1β)对其的保护作用及对热休克蛋白70(heat shock protrein70,HSP70)表达的影响。方法采用线栓法制作实验性RIRI大鼠模型。将32只Wistar大鼠随机分为正常组(1只)、假手术组(1只)、对照组(15只)、实验组(15只)。于再灌注开始时给予对照组大鼠玻璃体内注射平衡盐溶液5μL,实验组大鼠玻璃体内注射NRG-1β5μL,光镜下观察各组视网膜内层厚度及浸润视网膜的中性粒细胞数目、神经节细胞数变化;免疫组织化学方法观察各组视网膜内层组织中HSP70的表达情况。结果实验组RIRI后6h视网膜内层厚度均较对照组厚,早期视网膜内层水肿增厚,晚期视网膜神经节细胞数目减少及视神经纤维层萎缩变薄,神经节细胞数目多于对照组,而视网膜中的中性粒细胞数目少于对照组;HSP70于再灌注后24h达到高峰,但各时间段实验组表达强度均较对照组明显增强;实验组阳性细胞数在RIRI6h后各时间段较对照组显著增多(P<0.05)。结论NRG-1β对RIRI有保护作用;大鼠RIRI能诱导HSP70表达,NRG-1β能增强HSP70的表达。Objective To observe the effects ofneuregulin-1β(NRG-1β) on the retina and on the expression of heat shock protein 70 ( HSP 70) in rat' s retina after isehemia-reperfusion injury. Methods The rat model of experimental retinal isehemial-reperfusion was made by line occlusion. Thirty-two Wistar rats were divided into normal group ( 1 rat ), sham-operated group ( 1 rat ), control group( 15 rats) and experimental group( 15 rats). Five μL BSS was injected into vitreous cavity of control group and injected 5 μL Neuregulin-1β into experimental group as the same way. Retinal hietological changes were observed, the number of RGCs and the leukocyte infiltration in retina were counted and image analysis system was used to detect the thickness of inner retinal layer. The expression of HSP70 was assessed by immunohistochemistry. Results The inner layers of the retina of the experimental group was thicker than that of control group at 6 hours after retinal reperfusion injury. During the early period of reperfusion, the edema of retina was reduced in experimental group than that of control group, the number of RGCs of experimental group was more and the number of leukocyte was less than that of the control group. The expression of HSP70 redched the peak at 24 hours after retinal isehemia reperfusion injury, the expression of the experimental group was relatively obviously more than that of the control group. Positive cells of experimental group were more than that of control group at each time point after retinal isehemical reperfusion injury 6 hours. Conclusion NRG-1β could preserve retina after isehemia reperfusion injury,and enhance the expression of HSP70 ,which can also bc induced by retinal isehemial reperfusion injury.
