肿瘤病人血清中癌细胞凝集因子单克隆抗体的制备

摘  要:①目的通过制备癌细胞凝集因子的单克隆抗体(McAb),进一步研究该凝集因子的结构、功能、组织来源,并为临床建立检测试剂盒打下基础。②方法采用杂交瘤技术将免疫小鼠脾细胞与SP2/O细胞融合,经ELISA法结合癌细胞凝集抑制实验筛选出阳性克隆。③结果融合阳性率78.1%;筛选出的杂交瘤细胞染色体数目为96~106条,SP2/0骨髓瘤细胞为68条,小鼠脾细胞为40条,得到的McAb亚类分别是IgG1(E9)和IgG2α(E11).④结论获得了2株既能分泌抗体,又能阻断癌细胞凝集的杂交瘤细胞株(E9和E11)。Objective To prepare a hybridoma cell line which secret McAb against CCAF and can be used for further study of CCAFs chemical structure, function and tissue origin in order to provide theoretical data for preparation of laboratory kit. Methods Spleen cells from BALB/C mice immunized with CCAF were fused with SP 2/O myeloma cells. Positive clones were screened by ELISA combined with the test of inhibiting cancer cell aggregation. Results The fusion positive rate was 78.1%. The number of hybridoma cell chromosome was from 96 ̄106, The McAb subclasses were IgG 1(E 9) and IgG 2α (E 11 ), respectively. Conclusion The present experiment successfully obtained 2 strains of hybridoma cells which could either secrete McAb against CCAF or inhibit cancer cell agglutination.
