摘  要:目的研究小体积肝移植后受体来源的造血干细胞跨分化现象及影响因素。方法采用性别错配的大鼠不同体积肝脏移植,动态观察移植肝体积的大小和缺血损伤对术后骨髓干细胞向移植物内肝细胞跨分化促肝脏再生过程的影响。结果全肝移植组Y-染色体肝细胞阳性率为0.66%,而60%和30%肝移植组Y-染色体肝细胞阳性率分别增加到1.89%和3.78%,3组间差异均有显著性(P<0.05)。延长冷缺血时间使得肝内Y-染色体阳性肝细胞率减少到0.73%,与1h缺血时间组差异有显著性(P<0.05)。结论减体肝移植能增强HSCs在肝脏内向肝细胞跨分化。而延长冷缺血时间明显降低HSCs在肝内的跨分化。[Objective] To investigate the frequency of the transdifferentiation into hepatocytes of recipient-origi- nated hematopoitic stem cell (HSC) in different small-for-size fiver transplantations in rats and evaluate some factors influencing the recipient-derived cell repopulation during the fiver regeneration. [Methods] A sex-mismatched female to male small-for-size liver transplantation rat model was used to investigate recipient-derived HSCs transdif- ferentiation into hepatocytes. Fluorenscence in situ hybridization (FISH) was employed to demonstrate the Y-chromosome positive hepatocytes to analyze the effects of different grafted liver masses and ischemia damages on the frenquency of the recipient-derived HSC transdifferentiated hepatocytes. [Results] Compared with full liver transplantation (0.66%), the ratios of the Y-chromosome positive hepatocytes with all hepatocytes counted in 60% and 30% partially transplanted liver were prominendy increased to 1.89% and 3.78% respectively (P 〈0.01). The weights of livers in all groups reached about 50% on day 3 postoperation and about 100% on day 7 postoperation. In ischemia 6 hour group, the ratio of Y-chromosome positive hepatocytes was significantly lower than in the 1 hour group (0.73% versus 3.78%, P 〈0.05). [Conclusion] To some extend, the smaller the mass of partially resected fiver was, the higher frequency of HSCs transdifferentiate into hepatocytes in transplanted liver was. Prolonged cold ischemia time reduces the incidence of HSC transdifferentiation in grafted fiver.
