摘  要:以壳聚糖为手性添加剂,利用毛细管电泳对色氨酸对映体进行拆分,考察了壳聚糖浓度、缓冲液的pH和Cu2+浓度对分离过程的影响.实验结果表明,在壳聚糖的质量分数为0.02%、缓冲液的pH为3.5、Cu2+浓度为2mmol/L条件下,色氨酸对映体分离效果良好,分离因子为4.95,分离度为1.6.To establish a capillary electrophoresis (CE) method for the separation of tryptophan enantiomers,chitosan,a cationic natural polymer,was employed as a chiral additive selector. The separation was performed in untreated fused-silica capillaries using a Model CL1030 capillary electrophoresis system. Chitosan solution, ca. 0.01%- 0.20 % in 1% aqueous acetic acid was prepared as running buffer. As a complexing ion,Cu( Ⅱ ) ion concentration in the buffer was varied from 1 mmol/L to 4 mmol/L. The effects of the concentration of chitosan and Cu^2+ , pH of the buffer were investigated. A best separation of tryptophan enantiomers was achieved by using 0.02% chitosan(by weight) and 2 mmol/L Cu^2+ while the pH of the buffer was 3, 5. The separation factor was 4. 95 and the resolution was 1.6.
