芪归解毒汤防治硫酸镍致晶状体上皮细胞损伤的实验研究

摘　　要：目的观察芪归解毒汤药物血清对硫酸镍损伤正常人晶状体上皮细胞(Lens epithium cells,LEC,细胞编号:SRA01/04)的保护作用。方法采用血清药理学方法提取芪归解毒汤药物血清,体外培养晶状体上皮细胞,用硫酸镍产生细胞损伤,再加入体积分数分别为5%、10%、15%、20%的药物血清,倒置显微镜下观察晶状体上皮细胞的形态学改变,MTT法检测细胞活性的变化,试剂盒测量乳酸脱氢酶(LDH)含量的变化。结果倒置显微镜下观察到硫酸镍损伤的晶状体上皮细胞出现肿胀、体积缩小、折光性下降、贴壁率降低等形态学改变。加入10%和15%芪归解毒汤药物血清时细胞生长明显好转,折光性优于硫酸镍组,药物血清浓度为20%时细胞生长接近正常;MTT法检测药物血清组细胞活力较硫酸镍组明显增强;药物血清组LDH活性较硫酸镍组明显降低。结论芪归解毒汤药物血清能明显减轻晶状体上皮细胞的镍性损伤,并呈现剂量依赖关系。OBJECTIVE To study the effect of drag serum with Qigui Jiedu Decoction on damaged lens epithelial cells due to NiSO4. METHODS By using methods of serum pharmacology, lens epithelia cells were fostered in vitro and were divided into NiSO4 ＋ 5 % medicine group, NiSO4 ± 10 % medicine group, NiSO4 ＋15 % medicine group, NiSO4 ＋ 20 % medicine group, NiSO4 group and control group. Then the changes of cells in morphology, vitality in MTF and LDH were observed. RESULTS Qigui Jiedu Decoction showed protective function significantly on morphological change of lens epithelial cells, and could significantly strengthen the cell vitality and lower the LDH activity. CONCLUSIONS The drag serum with Qigui Jiedu Decoction could significantly alleviate the NiSO4 damage of lens epithelial.
