银杏叶注射液对兔慢性高眼压模型视网膜神经节细胞的保护作用

摘　　要：目的探讨银杏叶注射液对兔慢性高眼压模型视网膜神经节细胞的保护作用。方法采用2%α-糜蛋白酶注入家兔眼后房,造成实验性慢性高眼压。将造模成功的30只家兔随机分为三组,每组10只,分别给予以下治疗:静脉注射银杏叶注射液(银杏叶组)、银杏叶联合噻吗心安组(联合用药组)、不做任何治疗(模型对照组)。30天后各组分别进行视网膜神经节细胞凋亡检测正常节细胞计数和光镜观察视网膜各层病理改变和电镜观察神经节细胞超微结构的变化。结果每组均可观察到染色的阳性细胞,正常节细胞计数从多至少依次为联合用药组、银杏叶组和模型对照组(P<0.001),光镜下模型对照组视网膜各层结构最紊乱,联合用药组视网膜各层组织最完整。结论高眼压能导致视网膜神经节细胞发生凋亡,而银杏叶注射液能阻止其凋亡的发生,对视网膜神经节细胞有一定的保护作用。OBJECTIVE Study the protective effect of folium ginkgo injection on retinal ganglion cells in rabbit chronic high intraoeular pressure model.METHODS Injected 2 % α- chymotrypsin into rabbit posterior chamber to build up chronic high intraoeular pressure model.Thirty rabbits treated like that were separated randomly into three groups, each group with 10 rabbits, and then treated with different therapies as follows： Intravenous injection with folium ginkgo injection（folium ginkgo group）, folium ginkgo and Timolol treatment （combination therapy group）and control group. After 30 days of treatment, all groups were checked with retinal ganglion cells apoptosis, normal ganglion cells counting, pathological changes of retinal layers with light microscopy and ultra structure changes of retinal ganglion cells with electric microscopy. RESULTS Positive dying cells could be found in every group. Groups arranged with numbers of normal retinal ganglion cells decreasingly as following： combination therapy group, folium ginkgo group and control group（ P 〈 0.001 ）. In light microscopy, the worst arrangement of retinal layers is in control group, and the best is in combination therapy group. CONCLUSIONS High intraoeular pressure could lead to apoptosis of retinal ganglion cells. Folium ginkgo injection could prevent the apoptosis, which has the protective effect on retinal ganglion cells.
