胎盘多肽注射液对人脐静脉内皮细胞增殖的影响

摘　　要：目的:研究胎盘多肽注射液对人脐静脉内皮细胞(ECV-304)增殖的影响。方法:采用细胞计数和MTT法观察不同稀释度胎盘多肽作用72 h后,ECV-304细胞增殖情况。同时以免疫组织化学法检测不同稀释度胎盘多肽对ECV-304细胞增殖细胞核抗原(PCNA)表达的影响。结果:一定稀释度的胎盘多肽可促进ECV-304细胞的增殖,25 ml/L即可发挥作用,200 ml/L作用最明显。ECV-304细胞PCNA表达阳性率与胎盘多肽稀释度呈剂量相关性。结论:胎盘多肽可促进体外培养的ECV-304细胞增殖。Objective：To investigate the effects of placenta polypeptide on the proliferation of human umbilical vein endothelial cell（ECV-304） in vitro.Methods：ECV-304 cells were treated with different concentrations of placenta polypeptide,and the ECV-304 proliferation was observed with cytometry,and MTT in 72 h.Proliferating cell nuclear antigen（PCNA） was observed with immunohistochemistry method at the same time.Results：in the range of 25～400 ml/L,placenta polypeptide showed effect on ECV-304 proliferation in vitro in a dose-dependent manner and the maximal effect showed at 200 ml/L.Positive PCNA expression of ECV-304 correlated with placenta polypeptide concentrations used.Conclusion：Placenta polypeptide could promote the proliferation of ECV-304 in vitro.
