红鲫Sox1和Sox11b基因保守区的克隆与表达分析

摘　　要：目的进一步理解鱼类Sox基因的进化和表达方式。方法采用兼并引物PCR,从红鲫基因组DNA中克隆、测序和分析Sox基因。采用RT-PCR,研究Sox基因在红鲫13种不同组织中的表达。结果克隆和测序了红鲫Sox1-1、Sox1-2、Sox1-3、Sox11b1-1、Sox11b1-2和Sox11b2基因的HMG盒区序列。Sox1-1、Sox1-2和Sox1-3编码相同的氨基酸,在肝、脾、肾、心脏和脑组织中均有不同程度表达,在脑组织中检测到大量的转录物。Sox11b1-1和Sox11b1-2编码相同的氨基酸,系统发生树显示,红鲫Sox11b1和Sox11b2基因与斑马鱼Sox11b基因聚在一起,而远离斑马鱼Sox11a基因。在下丘脑和小脑中检测到Sox11b1-1和Sox11b1-2基因转录物,而没有检测到Sox11b2基因转录物。结论Sox1和Sox11b基因在进化过程中发生了复制。Sox1基因可能在神经系统中起着重要的作用。Sox11b1-1、Sox11b1-2和Sox11b2有相似而不同的表达方式。Objective To further understand the evolution and expression pattem of Sox genes in fish. Methods Genomic DNA from the red crucian carp was used as the template for PCR amplification using degenerate primers PCR. PCR products were cloned, sequenced, and analyzed. The expression patterns of Sox genes in 13 adult tissues of the red crucian carp were analyzed by RT- PCR. Results Six HMG- box sequences of Soxl - 1, Soxl - 2, Soxl - 3, Soxllbl - 1, Soxllbl - 2 and Soxllb2 were cloned and sequenced. Soxl - 1, Soxl - 2 and Soxl -3 shared the same amino acid sequences, and expressed unequally in the adult liver, kidney, spleen, heart and brain. High amount of transcripts were detected in the brain. Soxllbl - 1 and Soxl l bl -2 encoded the same amino acid sequences. The phylogenetic tree indicated that the red cmcian carp Soxllbl and Soxllb2 were clustered together with the zebrafish Soxl lb and were apart from the zebrafish Soxl l a. Transcripts of Soxl lbl - 1 and Soxl lbl - 2 genes were detected in the hypothalamus and cerebellum, where no transcripts of Soxl lb2 were detected. Conclusions Soxl and Soxl lb genes have been duplicated during the evolution. Soxl gene may play an impollant role in the nervous system. Soxllbl - 1, Soxl lbl -2 and $oxl lb2 have similar but distinct expression patterns
