人乳头瘤病毒11型E7蛋白检测尖锐湿疣患者宫颈分泌物抗体的意义

摘　　要：[目的]研究人乳头瘤病毒(HPV)11型E7蛋白在原核细胞的表达及检测尖锐湿疣(CA)患者宫颈分泌物中的抗体意义。[方法]将重组质粒pET32a(+)/HPV11E7经异丙基硫代-β-D-半乳糖苷(IPTG)诱导后表达,用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳和蛋白质印迹法(WB)分析鉴定;经镍螯合亲和层析胶体(Ni-NTA Agarose)纯化HPV11E7融合蛋白后作抗原,用间接ELISA法对106例CA患者、47例宫颈癌患者和98名健康对照者宫颈分泌物标本进行sIgA抗体检测。[结果]HPV11E7融合蛋白在原核表达系统中呈较高效的表达(浓度为40μg/ml),CA组、宫颈癌组和健康对照组宫颈分泌物sIgA抗体均值分别为0.12±0.01、0.06±0.005和0.05±0.007,阳性率分别为48.11%(51/106)、10.6%(5/47)和5.1%(5/98)。CA组宫颈分泌物sIgA抗体均值及阳性率均高于宫颈癌组和对照组(均P﹤0.01),对CA患者宫颈分泌物抗体检测的敏感度和特异度分别为48.11%和94.9%。[结论]HPV11E7融合蛋白具较强的抗原性,用于CA宫颈分泌物诊断试剂的研制具有应用价值。[Objective] To study the expression of the Human papillomavims （HPV） 11 type E7 protein gene in prokaryotic cells and its significance in detection of the antibody of the cervical secretion of eondyloma aeuminatum （CA） . [ Methods ] The expression products of His-E7 fusion protein were induced by IPTG, followed by analyzing with SDS-PAGE and Western blotting. The HPV11 E7 was purified with Ni-NTA Agarose affinity column to fuse the protein to be antigen, and the cervical secretion of eondyloma acuminatum among 106 patients with CA, 47 patients with cervix cancer and 98 healthy controls were detected the sIgA antibody by indirect ELISA method. [ Results] High expression of His-E7 fusion protein was detected in the prokaryotic expression system with the concentration of 40ug/ml. The values of slgA antibody in the cervical secretion of CA group, cervix cancer group and healthy controls were 0.12±0.01, 0.06 ±01005 and 0.05±0.007, respectively, and their positive rate were 48.11% （51/106）, 10.6% （5/47） and 5.1 （5/98）, respectively. When compared with the ten, ix cancer group and healthy controls, the positive rate of sIgA antibody in CA group was significant higher （P 〈 0.01 ）. The sensitivity and specificity of detection for sIgA antibody of patients with CA were 48.11% and 94.9 %, respectively. [ Conclusion] The HPV11 E7 fusion protein possessed strong antigenieity, which has application value to study CA cervical secretion diagnosis.
