摘  要:目的利用mRNA差异显示技术有效地比较组织或细胞间的差异表达基因.方法以玉米自交系B37Ht1与玉米大斑病菌0号及1号小种构成非亲和性互作与亲和性互作体系,利用mRNA差异显示技术对接种玉米大斑病菌后玉米叶片早期表达基因进行分析,寻找玉米抗大斑病相关基因表达片段.结果提取玉米总RNA;经引物筛选,获得8条随机引物;经聚丙烯酰胺凝胶电泳和银染检测,获得差异片段62条.结论利用mRNA差异显示技术能够获得玉米抗大斑病相关基因片段.Objective To effectively compare the differential expressed gene fragments with normal tissue or cells by mRNA differential display reverse transcription polymerase chain reaction (DDRT- PCR). Methods The gene fragments expressed differently in the corn leaves were studied and analysed both in incompatible combination (B37Htl inoculated with race 0 of Setosphaeria turcica) and in compatible combination (B37Htl inoculated with race 1 of Setosphaeria turcica) by mRNA differential display PCR. The amplification products were compared under the condition of background with one accord to obtain the gene fragments related to resistance to Setosphaeria turcia. Results 62 gene fragments expressed differently were obtained by PAGE.
