复方积雪草含药血清对肾小管上皮细胞Activin A和TGF-β1 mRNA表达的调节作用

摘　　要：目的:探讨复方积雪草对炎症因子刺激下肾小管上皮细胞生物活性改变的干预作用。方法:实验用IL-1β刺激肾小管上皮细胞,用不同剂量复方积雪草含药血清进行干预。用福辛普利(血管紧张素转换酶拮抗剂)含药血清作为对照。TGF-β1、Activin A和Activin A受体ActRⅠ等基因表达采用逆转录-聚合酶链式反应(RT-PCR)方法。结果:IL-1β诱导可使肾小管上皮细胞TGF-β1和Activin A的基因表达较正常对照细胞显著增高。各浓度复方积雪草含药血清干预均可下调Activin A及其受体的基因表达,但仅高浓度复方积雪草含药血清对TGF-β1的基因表达有显著下调作用。结论:复方积雪草对肾小管上皮细胞Activin A及其受体Act RⅠ的基因表达有下调作用,可能是其抑制炎症因子诱导的肾小管上皮细胞发生细胞活性改变,降低肾小管间质进一步炎症损伤的重要机制之一。福辛普利亦有相似作用,但机制可能不完全相同。Objective：To expore the effects of serum containing Centella Asiatica Compound on alteration of biological activity in renal tubular epithelial cell stimulated with IL - 1β by examining the mRNA expression of Activin A and Transforming growth factor β1 （TGF - β1 ）. Methods： Cultured mice primary tubular epithelial cells in vitro. Model group cells was stimulated by IL- 1β （10ng/mL）. The cells of CenteUa Asiatica Compound groups were co -incubated with serum containing Centella Asiatica Compound at same time. The cells without IL - 1β and Centella Asiatica Compound were regarded as control group. The mRNA expression of Activin A and its receptor Act R Ⅰ and TGF -β1 were assessed by reverse transcriptionpolymerase chain reaction analyses（ RT- PCR）. Results： The mRNA expression of Activin A and TGF -β1 in tubular epithelial cells stimulated with IL- 1β were significantly higher than normal group cells. Serum containing Centella Asiatica Compound could significantly reduce the expression of TGF-β1 ,Activin A and Act R I on lev- els of mRNA. Conclusion ：The role of Centella Asiatica Compound reducing mRNA expression of TGF-β1 and Activin A could be one of its important mechanisms of inhibiting the alteration of biological activity stimulated by inflammatory factor in renal tubular epithelial cell.
