摘  要:基于油菜纯合双显性雄性核不育恢复系的芯片表达谱分析结果,本文选择与糖代谢途径相关的两个基因GAPC和AT2G36580,采用荧光定量PCR方法对其在根、茎、叶、花、雌蕊、雄蕊、角果中的表达模式进行分析。结果表明,GAPC和AT2G36580在根、茎、叶、花和角果中均有表达,其中在根中表达量最高,花中表达量最低;GAPC在不育株花蕾发育的减数分裂至单核期被显著抑制,三核期表达量上升,而在可育株花蕾减数分裂至单核期表达量较高,四分体至单核期表达量最高(是减数分裂期的5.06倍),三核期表达量下降;AT2G36580在不育株花蕾发育中一直被抑制,但在可育株花蕾减数分裂至单核期表达量较高,四分体至单核期表达量最高(是减数分裂期的1.69倍),三核期表达量显著下降。进一步对雌雄蕊中的表达量进行分析表明,这两个基因均在可育植株雄蕊中表达量高,分别是不育株雌蕊(参照)的9.11倍和4.47倍,在雌蕊中表达量比参照显著低;与参照比,在不育株雌雄蕊中表达被显著抑制。初步结果表明GAPC和AT2G36580在油菜恢复系J10中具有相似的表达模式,可能是通过影响雄蕊发育过程中的糖代谢而参与花蕾发育的调控,与不育有密切关系。Gene expression profiles of GAPC and AT2G36580 related to the glycolysis metabolism pathway were detected by real-time quantitative PCR in a restorer J10 ( MsMsrfrf × MsMsRfrf) of dominant digenie male sterility of Brctssica napus. The results showed that the mRNA can be detected in roots, stems, leaves, flowers and pods,with the highest expression level in roots and the lowest in flowers. GAPC was inhibited in meiosis and uninu- cleate stages of sterile buds,but was highly expressed in trinucleate stage. In fertile buds GAPC hold the highest expression level at tetrad stage (5.06 times of that at the meiosis stage). AT2G36580 expression was inhibited in all bud developmental stages of sterile plants whereas there were highly expressed in meiosis and uninueleate stages of fertile buds. Further analysis of GAPC and AT2G36580 in stamens and pistils showed that expression level in stamens of fertile buds were 9.11 and 4.47 fold higher than that of sterife bads respectively. There were a significantly lower expression in pistils of fertile buds and a similar expression in stamens of sterile buds, in comparison with that in pistils of sterile buds. These data suggested similar expression profiles of GAPC and AT2G36580 in J10 may play an important role in sterility control via glyeolysis pathway.
