摘 要:目的运用生物信息学方法预测幽门螺杆菌分泌蛋白,为后续研究奠定基础。方法用Signalp和TMHMM两个软件对幽门螺杆菌全基因组开放读码框(ORFs)进行预测,Microsoft Access数据库分析处理以识别分泌蛋白,再与HpNCTC26695二维凝胶电泳质谱鉴定蛋白质组数据库(2DE-MS)和免疫蛋白质组研究结果进行比对验证。结果从Hp的1587个编码蛋白的ORFs中预测出191种分泌蛋白,其中27个预测蛋白与二维凝胶电泳质谱鉴定(2DE-MS)的Hp蛋白数据库中的蛋白一致;13个预测蛋白出现在高抗原性的免疫蛋白质组中。结论生物信息学方法可作为研究分泌蛋白的辅助工具,并可为研制Hp诊断试剂和疫苗提供候选抗原。To establish a whole genome prediction strategy for the secreted proteins from Hellicobacter pylori (Hp)in order to provide basis for the further studies on Hp,the open reading frame(ORF)of the whole genome of Hp NCTC 26695 was scanned by means of Signal P and TMHMM software,and the protein data analysis system based on Microsoft Access was established to process the output of these two software and to identify the secreted proteins. The predicted secreted proteins were verified with HpNCTC26695 two dimension electrophoresis followed by mass spectrometry(2DE-MS)database and immunoproteome of Hp. It was found that 191 secreted proteins were predicted from 1587 ORFs of Hp,in which 27 predicted secreted proteins were consistent with proteins in the protein database of Hp identified by 2DE-MS,and 13 of them were found to be highly antigenic in the immunoproteome of Hp. It is apparent that bioinformatics can be used as an assistant tool in the investigation of proteins and may provide candidate antigens for the development of diagnostic reagents and preparation of vaccines
