摘 要:背景:透明质酸和活性玻璃都是应用于创伤修复的生物材料,先前查寻的报道主要为二者单方面的实验结果。目的:拟验证不同配方活性玻璃与透明质酸复合凝胶在大鼠创面修复中的作用。设计、时间及地点:实验于2005-10/2006-03在解放军第二军医大学动物中心实验室完成。材料:SD大鼠60只,雌雄不拘,体质量300g左右;活性玻璃和透明质酸复合凝胶由上海鲁高生物科技公司制备提供,配方1为活性玻璃100%;配方2为活性玻璃95%,透明质酸5%;配方3为活性玻璃90%,透明质酸10%。方法:将SD大鼠30只,麻醉状态下,在脊柱两侧各1.5cm处环形全层切除皮肤,形成直径1.8cm的圆形皮肤缺损创面4个,共120个创面,采取双盲随机法,分别给予配方1,2,3及生理盐水对照组外用处理,每种方法处理30个创面。选取较优配方凝胶后,另取SD大鼠30只,用同样方法随机采取该配方或生理盐水外用处理。主要观察指标:分别于伤后1,2,3,5,7,10,12d观测平均愈合时间、创面面积、肉芽组织和上皮组织的增生情况、组织学切片。结果:①伤后第3天,配方2肉芽组织生长最先与周围组织相平且凸凹度较小;伤后7d,肉芽组织已基本充填创面。②镜下表现肉芽组织内密集成纤维细胞,新生毛细血管极其丰富,内皮细胞增多,胶原纤维也明显增多。配方1,2,3,与对照组之间差异均有显著性意义(P<0.05),以配方2最为显著,平均愈合时间也最短。结论:适当比例配方活性玻璃复合透明质酸能促进创面愈合。BACKGROUND: Hyaluronic acid (HA) and bioactive glass (BG) are both biological materials used in wound healing. Previous research mainly focuses on either HA or BG. OBJECTIVE: To explore effects of compoand gel used in rat wound healing, which is made by different proportions of HA and BG. DESIGN, TIME AND SETTING: The experiment was performed in the Central Laboratory, the Second Military Medical University of Chinese PLA (Shanghai, China) from October 2005 to March 2006. MATERIALS: Sixty SD rats were taken as test animals, of either gender and aged 300 g. The gel composite of BG and HA was provided by Shanghai Lugao Biological Technology Company (China). Formulation 1 contained BG only; Formulation 2 was made up of 95% BG and 5% HA; Formulation 3 contained 90% BG and 10% HA. METHODS: Thirty SD rats were anesthetized to make 4 round skin deficient wounds in each rat by cutting full-thickness skin on two sides of the spine, where was 1.5 cm away from posterior median line. Totally 120 wounds of 30 rats were treated by sodium chloride (control group) and formulation 1, 2, 3 of the gel, respectively. Each treatment dealt with 30 wounds. Other 30 SD rats were subjected to the optimal gel or external treatment of sodium chloride with the identical way. MAIN OUTCOME MEASURES: The average healing time, wound area, granulation tissue hyperplasia, epithelial tissue hyperplasia, and histological section were all estimated on the days 1, 2, 3, 5, 7, 10 and 12 after wound. RESULTS: Lamellar granulation growth was found on the third day. Granulation of the formulation 2 group was the first to reach the edge of wounds, and then fulfilled the wounds on the seventh day, Under microscope, it was found fibroblast concentration, endothelial cell hyperplasia, collagen fiber hyperplasia and blood capillary hyperplasia in the granulation.
