摘  要:以国产槲寄生为原料,研究了提取温度、料液比、提取时间和提取次数等因素对槲寄生多糖提取率的影响,并优化了槲寄生多糖中脱蛋白、脱色及醇沉技术.研究结果表明:提取温度95℃,料液比1∶26,提取时间3 h,可获得最佳的多糖提取率4.08%;之后对槲寄生粗多糖采用三氯乙酸法去除杂蛋白、活性炭法脱色,经80%乙醇沉淀,干燥后得槲寄生多糖产品.经紫外和红外光谱分析,国产精制槲寄生多糖不含蛋白和核酸,主要是吡喃多糖.Viscum coloratum ( Kom. ) Nakai is used as raw material, and the effects of extraction temperature, ratio of raw material to water, extraction time and extraction times on the yields of polysaccharides are studied through the single factor and orthogonal experiment. The technology of deproteintion, deolorization, and alcohol precipitation on Viscum coloratum ( Kom. ) Nakai polysaccharides ( CVPS ) are investigated. Results show that the maximal CVPS extraction ratio of 4. 08% can be realized when the samples are extracted using hot water as the extraction solvent under certain optimized conditions, i.e. the ratio of raw material to water of 1 : 26, the temperature of 95 ℃and the time of 3 hours. The CVPS is precipitated by 80% ethanol after eliminating the impurity protein with trichloroacetic acid (TCA) method and removing color with activated charcoal method. The product can then be obtained after desiccation. In addition, the ultraviolet spectrum and the infrared spectrum analysis show that a majority of CVPS is pyranopolysaccharides, and there is no protein and nucleic acid.
