栀子高频植株再生体系相关因子的优化

摘　　要：采用正交实验和单因子实验以及植物组织培养的方法,对栀子高频植株再生体系有关因子的进一步优化进行了探讨。结果表明,栀子带芽茎段再生体系的最佳消毒方式为70%酒精处理30 s后再用0.1%HgCl2处理15 min;栀子带芽茎段再生的最佳培养基为MS+6-BA 2 mg/L+NAA 0.5 mg/L;光照培养有利于栀子带芽茎段的再生;栀子带芽茎段再生的最佳蔗糖浓度为25 g/L,最佳琼脂浓度为7.5 g/L,最佳pH值为6,最佳培养温度为25℃。Objective： to illustrate the effects of several factors on dedifferentiation and redifferentiation of Gardenia jasminoides Ellis. earthnut. Method： orthogonal experiment, single factor experiment and tissue culture were applied. Result and Conclusion： the best disinfection way of Gardeniajasminoides Ellis. stems with a bud was firstly using 70% ethyl alcohol to process for 30 minutes then using 0.1%HgCl2 to process for 15 seconds again. The best regeneration culture medium of Gardeniajasminoides Ellis. stems with a bud was MS＋6-BA 2 mg/L ＋NAA 0.5 mg/L. The light culture was more advantageous to Gardenia jasminoides Ellis. stem regeneration. For regeneration system of Gardenia jasminoides Ellis. stems with a bud, the best concentration sucrose and agar was 25 g/L and 7.5 g/L respectively, the best pH value was 6, the best culture tempreture was 25℃.
