高效液相色谱法测定对硝基苯-β-D-葡糖苷的含量

摘　　要：目的:建立高效液相色谱法测定对硝基苯-β-D-葡糖苷的含量。方法:采用 Hypersil C_(18)色谱柱(250 mm×4.6 mm,5μm);紫外检测波长为303 nm;流础利为甲醇-水(30:70),流速为1 mL·min^(-1),柱温为室温。结果:对硝基苯-β-D-葡糖苷苷浓度在3.19～31.9 μg·mL^(-1)的范围内与峰面积具有良好的线性关系(r=0.9999);平均回收率为100.3%(n=9)。结论:本法简便、快速、准确,可用于对硝基苯-B-D-葡糖苷的含量测定。Objective ：To establish an HPLC method for determination of p -nitrophenyl -β- D -glucopyranoside （β- PNPG）. Methods ：β- PNPG was determinated by HPLC with a Hypersil C18 column （250 mm × 4. 6 mm,5μm） with UV detection at 303 nm, the mobile phase was contained of methanol - water （30： 70） , the flow rate was 1.0 mL ·min^-1, the column temperature was the room temperature. Results：The calibration curve was linear in the range of 3.19 - 31.9 μg·mL^-1（ r = 0. 9999） forβ - PNPG. The average recovery was 100. 3 % （ n = 9 ）. Conclusion：The method is simple, rapid, accurate and reliable,which may be used for the determination of β- PNPG.
