摘 要:为了探讨量子点(CdTe)诱导小鼠腹腔巨噬细胞(RAW264.7)凋亡及对线粒体膜电位的影响,用四甲基偶氮唑盐(MTT)法检测细胞增殖状况;Hoechst 染色进行细胞形态学观察;流式细胞术检测细胞凋亡及线粒体膜电位的变化.结果表明,小鼠腹腔巨噬细胞抑制率随着量子点作用时间延长和浓度增高而增加;形态学观察可见明显的凋亡特征;流式细胞仪检测结果显示剂量组细胞线粒体膜电位均较阴性对照组有所下降且细胞凋亡率显著增高,并呈现一定的剂量依赖关系,与阴性对照组比较差异有显著性(p<0.05).因此1.45~5.8μpg/mL剂量范围的量子点(CdTe)能抑制小鼠腹腔巨噬细胞的生长,导致细胞线粒体膜电位显著下降,引起小鼠腹腔巨噬细胞凋亡,从而认为这种凋亡可能是依赖线粒体途径的.To clarify the effect of quatum dots on apoptosis and mitochondrial membrane potential(MMP) in RAW264. 7 cells. MTT,flow cytometry (FCM) assays were performed to assess the cellular proliferation and apoptosis, and mitochondrial membrane potential respec- tively, and make cell Hoechst straining to observe the morphologic changes of apoptosis. Compared with the control, QDs inhibited cell proliferation in a dose and time-dependent manner. Hoechst 33258 images showed that most cells exposed to QDs exhibit apoptotic morphology change. After treatment of QDs, FCM assays showed the apoptotic rate of RAW264.7 cells were higher than those in the control (P〈0.05). Compared with control group, QDs caused a concomitant dissipation of the mitochondrial membrane potential, which showed obvious concentration-dependent relationship. QDs (1.45-5.8 μg/mL) could decreased the proliferative activity significantly, and induced RAW264. 7 cells apoptosis. The potential mechanism of RAW264.7 cells apoptosis induced by QDs might be related to the collapse of MMP.
