摘  要:研究了影响L-异亮氨酸(L-Isoleucine)产生菌钝齿棒杆菌(Corynebacterium crenatum)AS1.998原生质体形成和再生的各个因素。结果表明:菌体培养至对数生长期,0.8 U/mL青霉素G处理3 h后,1.0mg/mL的蛋清溶菌酶37℃,150 r/min酶解15 h,原生质体形成率可达98.0%以上,在含8.5%蔗糖的再生完全培养基上,再生率可达20.0%~30.0%。In order to optimize the conditions of protoplast formation and regeneration of Isoleucine-producing Strains Corynebacterium crenatum AS1. 998. The effects of different factors, such as penicillin, lysozyme dosage, temperature of lysozyme hydrolysis, time of lysozyme hydrolysis, rotation speed on protoplast formation and regeneration of Corynebacterium crenatum AS1. 998 were studied. The results showed: Bacterial of logarithmic culture pretreated with 0.8U/mL penicillin G for 3h is suspended in osmotic stabilizer and incubated with 1.0mg/mL egg-white lysozyme for 15h,the protoplast formation rate is above 98.0% and the regeneration rate is about 20.0%-30.0% in HCM containing 8.5% sucrose.
