摘  要:目的:探讨基质金属蛋白酶10(MMP10)基因及其蛋白在早孕小鼠子宫内膜组织中的表达规律及意义.方法:采用RT-PCR、原位杂交、免疫组化和Western-blot法分别检测受孕3-7 d的小鼠子宫内膜MMP10基因和蛋白的表达及分布,并以未孕小鼠子宫内膜为对照.结果:①早孕小鼠子宫内膜组织MMP10基因和蛋白的表达较未孕小鼠均显著增加,且表达量从着床前期到着床后期逐渐增强,其中d6、d7表达最强,与其他妊娠各期比较差异有统计学意义;②MMP10基因和蛋白表达主要分布在子宫内膜间质细胞.结论:MMP10在早孕小鼠子宫内膜组织中高表达可能有利于降解细胞外基质,促进胚胎滋养细胞的侵袭,参与着床中子宫内膜的蜕膜化过程以及胚胎的早期增殖与分化.Objection: To explore the expression rule of MMP10 in endometria of early pregnant mouse. Methods: RT-PCR, in-situ hybridization, immunohistochemistry and Western blotting were used to detect the MMP10 gene and protein in the mouse endometria from pregnant d3, d4,d5, d6 and dT, as in contrast with non-pregnant(d0) endometrium. Results: The MMP10 expression in the pregnant groups was higher than that in the non-pregnant group, and the MMP10 expression increased significantly in the postimplantation (d6 and d7). In-situ hybridization and immunohistochemistry showed that MMP10 expression of stroma cells in endometria was higher than that of uterine epithelium cells. Conclusion: The higher expres- sion of the MMP10 in endometrial stroma cells of early pregnant mouse may facilitate the implantation, proliferation and differentiation of embryos.
