负载冻融抗原的DC诱导特异性CTL杀伤胃癌细胞的实验研究

摘　　要：目的研究负载肿瘤抗原的树突状细胞(DC)活化的特异性细胞毒性T淋巴细胞(CTLs)对胃癌细胞的体外杀伤作用。方法冻融法获取胃癌细胞抗原,联合应用粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)、白细胞介素4(IL-4)和肿瘤坏死因子α(TNF-α)诱导培养外周血DC并负载肿瘤抗原,激活自体T淋巴细胞,制备特异性CTLs,MTT法检测对胃癌细胞的体外杀伤作用。ELISA法检测γ干扰素(IFNγ)分泌情况。结果负载胃癌抗原的DC激活的CTLs表现出对胃癌细胞的特异性杀伤作用,产生高水平的IFNγ(P<0.01);而对B16黑色素瘤细胞没有杀伤作用不产生高水平的IFNγ。未负载胃癌抗原DC刺激的CTLs对胃癌细胞无杀伤作用。结论应用细胞因子从人外周血中诱导的DC负载胃癌抗原后,激活的CTLs在体外对胃癌细胞能产生高效而特异性的杀伤作用。Objective To investigate the antitumor efficiency of the special cytotoxic T lymphocytes（CTLs） activated by clendritic cells （DC） loaded with gastric cancer antigens ex vito. Methods Gastric cancer antigens were extracted by thawing and freezing. The mononuclear cells isolated from peripheral blood of gastric cancer patients were cultured in RPMI medium 1640 containing rhGM-CSF, IL-4, and TNF-α to generate dendritic cells. After pulsed in vitro with gastric cancer lysates, DCs were used to induce T lymphocytes into cytotoxic T lymphocytes for gastric cancer cells. The cytotoxicity of CTLs to gastric cancer cells was assayed by MTT. IFNγ production was determined with ELISA kit. Results CTLs stimulated with DCs loaded gastric cancer antigen showed the specific kill effect on gastric cancer cells, produced high levels of IFNγ. However, it did not showed specific kill effect on B16 melanoma cells and did not produce high levels of IFNγ. CTLs stimulated with DCs unloaded gastric cancer antigen did not show the specific kill effect on gastric cancer cells. Conclusion The antigen specific CTLs killed gastric cancer cells efficitively.
