摘  要:目的不同发育阶段的骨髓间充质干细胞性质会发生一些变化,对干细胞移植治疗产生影响,本研究试图分析不同年龄段人骨髓间充质干细胞的生长和增生情况,探讨年龄对于骨髓间充质干细胞的影响。方法采集61例不同年龄人的骨髓,分为少年组、中青年组和老年组,分离间充质干细胞,进行体外培养,观察其形态变化,对比分析其首次传代和每次传代时间、细胞克隆形成数量、细胞增长曲线的变化情况。结果体外培养的干细胞在形态上无年龄差异,均成梭形纤维状,早期呈克隆状生长。细胞的首次传代时间从少年到老年分别为:(12.3±5.1)d,(13.6±5.8)d和(16.7±7.2)d,呈逐渐增加的趋势,老年组明显长于另外两组;3组的体外培养倍增时间无明显差别,细胞克隆数量从少年组到老年组分别为:(32.3±3.1),(26.4±4.8),(18.8±5.0)个,老年组明显减少,老年组干细胞的增殖曲线也低于另外两组。结论人骨髓间充质干细胞的生长增生能力随着增龄逐渐减退,在老年组呈现出较明显的变化。Objective To analyze the growth and proliferation of bone marrow mesenchymal stem cells (BMSC) in people of different ages and to explore the effect of age on BMSC. Methods BMSCs were isolated from 61 persons of different ages, who were divided into 3 groups: teenage, middle age, and old age. The cells were cultured in vitro and different measures were carried out to observe their proliferation. Results There were no visual differences in the shape and size of the first generation between the 3 groups, but the BMSC of old people showed a little faster loss of spindle-formed features over cultivation and needed longer time to reach the first confluence than other 2 groups. The average time of reaching first confluence in teenage, middle age and old age groups was ( 12.3 ± 5. 1 ) d, ( 13. 6 ± 5.8 ) d and ( 16.7 ± 7.2) d, respectively. Doubling time during the following passages revealed no difference. Total CFU numbers of BMSC decreased with aging.. 32. 3± 3. 1,26.4 ±4.8 and 18. 8 ±5.0, and proliferation curve of BMSC was lower in old people group than in the other 2 groups. Conclusion Human BMSC experiences a gradual decrease in proliferation and replication ability with aging, which is more obvious in old people.
