摘 要:目的建立人单核源性泡沫细胞的疾病模型,为动脉粥样硬化(AS)的发生发展机制及其防治研究提供新途径。方法采用密度梯度离心法分离冠心病患者外周血单核细胞,以80nmol/L佛波酯(PMA)诱导48h使之转化为巨噬细胞,然后与80mg/L氧化低密度脂蛋白(ox-LDL)共孵育48h,使之转化为泡沫细胞。结果经PMA诱导后,光镜下观察发现单核细胞已转化为形态典型的巨噬细胞。与ox-LDL共孵育48h后,油红O染色和透射电镜观察发现细胞内出现了大量的红染颗粒或脂质空泡。高效液相色谱检测发现细胞内胆固醇(TC)明显增加,其中胆固醇酯(CE)含量大于TC的60%,符合泡沫细胞的定义。结论从离体细胞培养方面探寻并建立了人单核源性泡沫细胞的疾病模型,该模型的病生特性与人类AS疾病接近,具有制备方法简易、细胞材料来源广等优点。ObjeCtive To establish a human monocyte-derived foam cell model,so as to offer a novel method for studying the pathogenesis and progression mechanisms as well as the prevention and treatment of atherosclerosis (AS). Methods Monocytes were isolated from peripheral blood of coronary heart disease (CHD) cases by Ficoll-Hypaque density gradient centrifugation. The isolated cells were induced by phorbol 12-myristate 13-acetate (PMA) at 80 nmol/L for 48 h and coincubated with ox-LDL at 80 mg/L for another 48 11. Results Macrophages in typical morphologies were successfully transferred from monocytes induced by PMA. When incubated with 80 mg/L ox-LDL,the ratio of cellular cholesteryl ester to total cholesterol in macrophages was beyond 60% identified by HPLC analysis,and there were also many lipid droplets displayed in cells beth under transmission electron and light microscopes, which indicating the formation of foam cells. Conclusions A foam cell model is successfully established in vitro from monocytes of CHD cases. The physiopathologic characteristics of the model are some more similar to that of human AS diseases than other models, further more, the derived ceils are easily obtained and the model is easily made.
