摘 要:目的:研究神经组织定位材料经不同染色后在玻璃切片上粘附能力,探索一种适合臂丛神经组织切片三维重建的定位材料。方法:将臂丛神经分别与人发、尼龙线(11-0),锦纶线(6-0)、铂丝(20μm)4种材料用OCT包埋固定于专用固定架,然后进行冰冻组织切片。分别用普通玻片和多聚赖氨酸涂层玻片进行贴片。然后分别进行乙酰胆碱酯酶(ACHE)组织化学染色法和常规HE染色,最后在显微镜下记录染色前后不同材料在玻片上残留的定位点的数目,比较4种不同材料在不同玻片上的粘附结果。结果:在以上4种材料中,采用乙酰胆碱酯酶(ACHE)组织化学染色比HE染色能够更少地减少定位点的丢失。采用多聚赖氨酸涂层的方法比普通玻璃切片能够具有更好的组织粘附力。人发在采用乙酰胆碱酯酶(ACHE)染色时,使用多聚赖氨酸涂层玻片可以达到最好的粘附效果。结论:在以上4种材料中,人发是较为理想的臂丛神经组织切片三维重建的定位材料。Objective:To study the adhesion ability of different nerve locating materials on the slices after using different kinds of staining methods and to find an ideal locating material for three-dimensional reconstruction of human brachial plexus.Methods:Four materials [the hair,nylon thread(11-0),nylon line(6-0),platinum wire(20 μm)] were embedded with brachial plexus using OCT as embedding material and fixed in exclusive holder separately.Then serial nerve tissue frozen section were made.Nerve sections were adhered to ordinary glass and poly-L-lysine coated glass respectively.Continuously,acetylcholinesterase(ACHE) staining and conventional HE staining was made separately.At last,remnants of the number of locating points were recorded under the microscope and four different materials in different slides adhesion results were compared.Results:The use of acetylcholinesterase(ACHE) staining method can be less loss of locating points as compared with routine HE staining method in the above four materials and use of poly-L-lysine coated glass slicing could get better adhesion ability than ordinary glass.Hair could achieve the best adhesion effect by the use of poly-L-lysine coated glass after acetylcholinesterase(ACHE) staining.Conclusion:The hair is a relatively ideal locating material for the three-dimensional reconstruction of brachial plexus in the above four materials.
