摘 要:【目的】寻找与番茄抗叶霉病基因Cf6连锁的分子标记。【方法】以番茄抗叶霉病和感叶霉病亲本组合(03036×03748)的F2分离群体为研究材料,采用BSA法和RAPD技术筛选与抗病基因连锁的分子标记。【结果】经F2单株分析,在后代群体中扩增出了两条长约500bp的特异片段,该特异标记与叶霉病抗性基因Cf6紧密连锁,遗传距离为8.7cm。测序结果显示,目标片段的大小分别为619bp和559bp,并将该标记转换为SCAR标记。【结论】SCAR标记,可用于番茄叶霉病抗性分子标记辅助育种。[Objectivel The study identified the molecular marker linked to Fulvia fulva resistant gene Cf6 in tomato. [ Method] With a F2 population between a resistant and a susceptible parent (03036 ×03748), the linked marker was screened by BSA method and RAPD technology. [Result] Two specific bands were found at the 500 bp region using F2-based bulked segregant analysis plants. An RAPD marker was tightly linked to Fulviafulva resistant gene Cf6 in tomato, with a genetic distance of 8.7 cm, The RAPD marker was converted into SCAR marker. Sequencing of the fragment indicated that the length was 619 bp and 559 bp, respectively. [Conclusion] SCAR markers could be applied to identify the resistance to Fulviafulva, and can be used in molecular marker-assisted breeding practice.
