雷公藤多甙抗大鼠原位肝移植急性排斥反应研究

摘　　要：目的探讨雷公藤多甙的免疫抑制作用，以及其诱导免疫耐受作用。方法该实验应用肝移植120只SD大鼠，分为四组：对照组（A组），TII组（B组），CsA组（C组），TII＋CsA组（D组）。分别于术后1、3、5、7、14d检测外周血的IL-2R的含量和脾脏淋巴细胞IL-2、IL-10的活性表达及脾脏组织CD4^＋、CD25^＋、CD8^＋、CD28^＋的蛋白表达。结果三组治疗组的IL-2活性和IL-2R含量均明显低于对照组。三组治疗组问的比较是D组低于B、C组；三组治疗组的IL-10活性明显高于对照组，三组治疗组也有显著差异。即D组高于C组，C组高于B组，上述均有统计学意义（P〈0．05）。三组治疗组的CD4^＋、CD25^＋、CD8^＋、CD28^＋的蛋白活性表达均高于对照组，而CD28^＋的蛋白活性表达却相反。三组治疗组间的CD4^＋、CD28^＋的比较，均有显著性差异（P〈0．05）。结论雷公藤多甙不但具有免疫抑制作用，而且还能够诱导一定程度的免疫耐受。Objective To investigate the effects of GTW on acute rejection in rats after orthotopic liver transplantation （OLT）. Methods A total of 120 SD rats receiving OLT were divided into 4 groups： control group （A）, TII group （B）, CsA group （C）, TII＋CsA group （D）. Activity of IL-2R in peripheral blood, CD4^＋ , CD25^＋ , CD8^＋ , CD28^＋ in spleen and IL-2, IL-10 in spleen lymph cells were respectively detected on the 1st, 3rd, 5th, 7th and 14th d after operation. Results The serum levels of IL-2R and activity of IL-2 in the 3 treated groups were significantly lower than those in the control group. Among the treated groups, it was lower in D than in B and C. The activity of IL-10 in the 3 treated groups were significantly higher than in that in control group. Meanwhile, there were differences among the 3 treated groups： it was higher in D than in C and higher in C than in B. The activity of CD4^＋ , CD25^＋ and CD8^＋ in the 3 treated groups was higher than that in the control group. However, the activity of CD28^＋ was lower in the 3 treated groups than in the control group. There was significant difference in activity of CD4^＋ and CD28^＋ among the 3 treated groups. Conclusion GTW has the function of inducingimmune tolerance to some extent except for immune suppression.
