摘　　要：从制备的10种抗HCG单抗中筛选出B12－G4（抗a－HCG）和C10－A10（抗β-HCG）两种。含单抗腹水用45％硫酸按沉淀两次，经0．1mol／LpH2．0甘氨酸缓冲液预处理后，对聚苯乙烯胶乳进行物理吸附。致敬胶乳用于尿液HCG直接凝集试验，敏感度达0．3IU／ml．测定中加入了研制成的非特异性盛集抑制剂，稍除了假阳性反应．用本法测定了临床上50名妇女的尿液标本，并与胶乳凝集抑制法及免疫金渗滤法进行比较，其中49份标本三者的结果一致，1份标本本法与免疫金渗滤法是阳性，而凝集抑制法呈阴性。B12-G4 (anti-HCG-Q subunit) and C10-A10 (anti-HCG-βsubunit) were selected from ten anti-HCG monoclonal antibodies. The monoclonal antibodies contained in ascites were precipitated twice by 45% ammonium sulfate and treated with 0. 1 mol/L pH 2. 0 glycine buffer, and then absorbed onto the latex. The sensitized latex was used in the direct agglutination test of urine HCG, the sensitivity reached 0. 3 IU/ml.A nonspecific agglutination inhibitor was developed and appiied in the assay to eliminate the false positive results.50 clinical urine samples of women were tested by this method in comparison with those of latex agglutination inhibition test and immunogold filtration assay. 49 samples showed same results for the three methods, but in one sample, our mpthod and the immuno-gold filtration assay gave positive result while the agglutination inhibitor test gave negative result.
