摘  要:研究IL-1β、TNFα和G-CSF等细胞因子对裂变中子和γ射线照射小鼠的辐射防护作用,探讨其作用机理。方法观察几种细胞因子对受照小鼠存活率的影响,计数受照小鼠脾脏CFU-S,进行骨髓红系、粒系及混合系造血祖细胞培养,检测部分动物的血清GM-CSF水平及脾脏淋巴细胞转化能力的变化。结果IL-1β和TNFα能明显提高8.5Gyγ射线照射小鼠的30天存活率,二者复合应用有加强作用;裂变中子照射时防护效价下降,复合应用亦无加强作用。G-CSF也能提高中子照射小鼠的30天存活率。3种细胞因子单独或复合应用对受照小鼠的造血和免疫系统,特别是粒系造血有某种程度的保护作用。结论IL-1β、TNFα单独或复合应用对γ射线照射小鼠有明显的保护作用,裂变中子照射时的防护效价降低,且无复合应用的加强作用。G-CSF对中子照射小鼠有辐射保护作用。这些细胞因子的辐射防护作用可能与其减轻小鼠造血和免疫系统损伤和促进其恢复有关。Objective\ The radioprotective effects of cytokines,IL 1 β,TNF α and G CSF,on fission neutron or γ ray irradiated mice were investigated and their protective mechanisms were explored.Methods\ The 30 day survival rate of mice was determined.The numbers of CFU S and BM progenitor cells of mice were estimated.The levels of serum GM CSF and the change of transformation function of murine splenic lymphocytes were examined.Results\ IL 1 β or TNF α could obviously increase 30 day survival rate of 8.5 Gy γ ray irradiated mice.Administration of IL 1 β plus TNF α possessed synergistic effects.However,radioprotective effects of IL 1 β or TNF α decreased in neutron irradiated mice.Simultaneous injection of IL 1 β and TNF α did not possess additive effects,G CSF could also increase 30 day survival rate of fission neutron irradiated mice.Administration of IL 1 β,TNF α and G CSF alone or combinedly could protect the hemopoietic and immunological functions of mice exposed to neutron or γ ray radiation to a certain extent,especially protect granulopoiesis.Conclusion\ Administration of IL 1 β,TNF α alone or simultaneously could protect mice from γ ray irradiation obviously,their radioprotective effect on fission neutron irradiated mice reduced,and there were no additive effects. G CSF could protect mice from neutron irradiation.The mechanisms of the radioprotective actions of these cytokines might be related to their ability to alleviate the radiation injuries and promote the recovery of hemopoietic and immunological function of mice after radiation.
