免疫金银染色法测定抗中性粒细胞胞浆抗体

摘　　要：目的建立检测抗中性粒细胞胞浆抗体(ANCA)的免疫金银染色法,并评价其应用价值。方法以中性粒细胞基质片为抗原,以胶体金标记的羊抗人IgG为第二抗体,加银染液,普通光镜镜检判断结果;用此法和间接免疫荧光法(ⅡF)检测28例正常人和69例相关疾病的患者血清标本共97例。结果两方法的敏感性和特异性无差异,两法符合率为100%。结论免疫金银染色法可替代ⅡF法,便于基层单位应用。Objective To establish an immunogold silver stainining （ IGSS ） method for detecting anti- neutrophil cytoplasmic antibody （ANCA） , and to appraises its application value. Methods Take the neutrophilic granulocyte as the antigen and take the gold labeled sheep-anti-human IgG as the second antibody, then adds the silver colouring agent, judging the results by ordinary microscope ; the serum of 28 normal persons and 69 patients of correlation diseases were examined by IGSS and indirect immunofluorescence （IIF）. Results There is no obvious differences of the sensitivity and the specificity between the two methods and the coincidence rate is 100%. Conclusion The immunogold silver stainining （IGSS） can be substituted for IIF and is more suitable for the basic hospitals.
