摘 要:为建立草木樨的细胞融合原生质体杂交体系,以白花草木樨嫩叶为材料,以MS为基本培养基,在2,4-D和6-BA不同激素水平下研究愈伤组织诱导率,并在不同的酶液组分和酶解时间下对愈伤组织进行酶解制取原生质体。结果表明:随着激素2,4-D和6-BA浓度的增大,愈伤组织的诱导率呈先增大后减少的趋势变化,当激素2,4-D和6-BA浓度分别为1.5、0.3 mg/L时,诱导率最高,可达90%。草木樨愈伤组织在2.0%纤维素酶、0.3%果胶酶、1.0%离析酶中酶解15 h为最佳,此时有活力的原生质体产量最高,可达1 890×104个/g。In order to set up the system of cell fusion and protoplast hybridization, the leaves of Melilotus alum were used to induce the calli with MS under different hormones. And then the callus enzymolysis were carried out in different enzyme systems and different enzymolysis time. The results indicated that with the concentration of 2,4 -D and 6 -BA increasing, the callus inductivity increased firsdy and then declined. The ratio of callus formation was the highest of 90% in the culture medium with 1.5 mg/L 2,4 - D and 0. 3 mg/L 6 - BA. The optimum condition for preparing protoplast was the enzyme mixture of 2.0% Cellulase, 1.0% Macerozyme and 0.3% Pectinase and enzymolysis for 15 hours;under these conditions, the vital protoplast production was the highest, which could be 1 890× 10^4 individual per gram.
